If you have plaster board on battens then it's a different ball game - need to locate a batten at least. Once it has been turned with the gauge, the wings on the back of the fixing will rotate out and lock, which secures it firmly in the plasterboard.

Now use the gauge supplied to check that the fixing is either 3mm or 6mm below the surface (6mm for rendered plasterboard, 3mm for not). I was somewhat cautious about drilling into the cinder blocks as they seemed rather soft and crumbly. If you are fixing timber on without removing a section you can use the knocking technique to locate the studs.
